Isolation and Identification of Bacteria Causing Bent - neck of Cut Rose



Abstract
In cut rose, Rosa hybrida ¢¥Norena¢¥, microorganisms which cause vascular blockage were observed in the xylem vessels of stem ends after 10days in distilled water with light microscope, but not observed in freshly harvested cut rose stems. These microorganisms were isolated from the stem ends of cut rose, and these microorganisms were idientified as bacteria, Pseudomouas fluorescens, Klebsiella oxytoca and Aeromonas hydrophila by a transmission electron microscopy, physiological, biochemical and nutritional characteristics. When all the three bacteria were inoculated into holding distilled water to 10^5 cfu¡¤mL^(-1), the percentage of bent-neck was greatly higher than when one or two bacteria were inoculated. When myclobutanil, a triazole fungicide, was added into the holding distilled water inoculated with three kinds of bacteria isolated, the percentage of bent-neck greatly decreased in 6-8 §·¡¤L^(-1) of the fungicide than others.
